Abstract: Optical microscopy has undergone a renaissance over the past few years, enabling researchers to look deeper and with higher resolution than ever before. After presenting an overview of the limitations of standard microscopy, I will describe new techniques that go beyond these limitations. In particular, I will describe techniques involving nonlinear optical interactions, such as two-photon microscopy and second harmonic microscopy, which allow significantly increased depth penetration in thick tissue. I will also describe techniques involving structured illumination which provide high resolution beyond the standard diffraction limit. The goal of my presentation is to present an overview of the state-of-the-art of optical microscopy and to summarize some recent trends in the field.

Note: Jerome Mertz received an AB in physics from Princeton University in 1984, and a PhD in quantum optics from UC Santa Barbara and the University of Paris VI in 1991. Following postdoctoral studies at the University of Konstanz, Germany (Jürgen Mlynek group) and at Cornell University (Watt Webb group), he obtained a lecturer position at the Ecole Supérieure de Physique et de Chimie Industrielle in Paris, where he became a CNRS research director. He is currently an associate professor of Biomedical Engineering at Boston University. His interests are in the development and applications of novel optical microscopy techniques for biological imaging.
